Hi Sharma,
Assignments on Lookup main field is not supported from the primary main table.
Though the lookup main field is shown in the Assignment field drop down, the expression will always fail.
For any lookup field assignment, we can assign only a value which is existing in the lookup table which will be selected from the tab "Lookups" in the Assignment expression editable window.
The lookups tab does not show the lookup main table which means there is no possibility to select a valid value and assign it to the field.

Hi Sharma,
From Main Table it is not possible to assign values in the lookup field which is not maintained in corresponding lookup table.
Same is the case with Lookup[Main] also.First value should be assigned in the Looup tables and then main table looks into the value.
But yeah you can use Lookup[Main] table fields in the expression to check some condition.
Now you must be wondering when its not possible why we get Lookup[Main] and Lookup fields to select in the Assignment field.
There can be some situation where whenever a record is created default value should be set automaticaly in the lookup field.
But this case is not applicable with Lookup Main as explained by Sravan it is not available in the Lookup List in the expression editor.
