In terms of SAP SD testing - you can use test tools, when your test data and scenarios have stabilised a fair bit, otherwise, it would be better to use manual testing, considering that SAP SD interacts with a lot of other modules.
The tools used can range from SAP's own : eCATT to HP QC for updating test scripts & Test results. There is also a specific SAP testing tool, which is becoming popular - SAP TAO.
And as mentioned above, you can also use QTP to run scripts.

As for as my experience with testing i have done manual testing.
According to test cases test scripts will be prepared and out put will be reported to heads.
Usually CATT(computer aided testing tool) will be used.
For the more information pertinent to CATT you please google it. You will get immense information

As for as automation testing is concerned, the most widely used tool is Mercury 9.0.Apart from it SAS testing tool is also used.
Also to mention that for automation testing purpose, QTP script is used (its not testing tool, its a programming language to guide the computer how to test and what data is filled).
