Hi,
In SAP EP6 it is possible to use an SAP system as the persistent data store for the UME.
Is it also possible to use more than one R/3 system as a data store and use the combined users from those systems in the portal?

Yes. Assuming that you are on SP2, you will find the relevant documentation at:

No, you can use exactly one system (to be more precisely: one client of a system).
