You can set the spec in the material specification, but for a characteristic set up as a calculated characteristic, you must specify the formula in the inspection plan. I.e. You can't use calculated characteristics without an inspection plan.
FF :- From your reply I understand that we can not use Formula MIC (activated in Control Indicator) in Material Specification (QS61). We can only use Formula of MIC with Task List i.e QP01.

My Understanding :- When we assign a Formula based MIC in Tasklist, we get a space where in we define the formula in Tasklist itself. But when we assign the same Material Specification, system does not provide any space..?

Exactly you can not put formula in the Material specs !! as there is no provision to enter formula.
Ultimately you have to use same MIC in the task list.
