Different start dates for multiple locations

My company has a payroll start date of Monday, ending on Sunday. One of our locations would like to move the payroll start date back to Saturday for pay purposes to accommodate a different work schedule.

Are there implications to the cost of the program or ease of use? What would doing this require?

What you seem to require is a new Payroll Accounting Area with it's own Payroll Calendar.

I don't believe that it should "cost" you any more as far a your SAP Licence is concerned, but the additional costs would be in processing that PAA. That "location" would require to have it's own Personnel Area or Personnel Sub-Area, so a lot of configuration is involved, some additional annual maintenance (generating Payroll Calendars), and the process of transfering an employee between that location and an other one would require added vigilance (timing of the Action in relation to Payroll Calculation).
