It's an interesting question. I haven't seen many clients use a virtual system (i.e a placeholder) as a source system, but maybe I'm not fully understanding. How/where was the transport created that you're now trying to release?
If you're able to create a transport (assuming we're talking about a client on VMWare?) that has all the necessary software installed and set up as a managed system on Solution Manager, you should be able to release it as well.
I'll be interested to hear other views - one can always learn something new.
