If it is just about generating time periods for the time profile, yes you can update it post planning area activation. But if you are referring to add a new time bucket or remove an existing time bucket, then the answer is no.
In my opinion yes if you are not adding any more attribute to time profile than yes. I hope you chose the option to down load time profile with existing period . What exactly you want to do when you mention updating time profile
