Set the Automatic Customisation & Prefix numbers in SWU3 in Production ?

Dear Experts,

1. Do i need to set the Prefix numbers in SWU3 in Production system directly?

2. Do i need to do Automatic Customisation of SWU3 directly in Production?

I transported the requests but it has no effect,still they are in red cross in Production.
