If I have a formatted search in a User-Defined Field for an Item, will it update/refresh when I lookup that Item with the DI API?

only when you program it yourself, forcing a formatted seatch from di is not possible.

I will NOT update when you use the DI API, formatted searches are only active in the SBO Client.
