Will on-prem ECC applications be able to connect to an SAP ECC running on the AWS cloud using an on-premise PO system?

Yes, absolutely possible.

I have an environment where my PI/PO server running on premise in a different network zone and it's connected with my SAP instances running on AWS. You need to have VPC setup at AWS side, then you will have direct connection.

Moreover, let's assume you have another instance on a third network zone which is not a part of your VPC then you can simply use SAP router to build a secure connection to your AWS systems.
