Not such a simple question. Define "single instance"?

Same user same system/browser, same user different systems, same system different users, different users different systems?

Then use any locking/tracking mechanism you like to catch the scenario you're trying to prevent. Lock objects, Z-table, cookies, ...

Then also consider that sessions are not 'connected' as such, so WD cannot detect what the user is doing. When the user goes to lunch have they closed the browser? Are they starting again or do they still have another tab open?

So the answer is even more complex than the question. Generally a WD app should be designed as if it were stateless in order to avoid this need in first place.

Thank you for your answer,

single instance was meant to be that at the same time, application can be used just by one user, in one browser and on one system.

OK, so same answer is still relevant, just make sure that whatever solution you go for applies system-wide.
