Yes, But it depends upon the scenario.
If both the receiver receives the message of same message type then u can use only one mapping itself... but still u can use two message mapping but it is meaning less to have a two different message mappings for the same source and target message type. So u can go for multiple mappings when the one mapping requirement differ from eachother.

Yes you can ...
Create different Interface mappings for diffenent mappings , like one for Message Mapping other for ABAP mapping and use these Interface mappings in the Interface Determination for those two receivers.
