Hi,
if the RFC communicatoin is syncronouse mode, do we need to create two RFC destinations in R3 , XI aswell?

I think you have confused because of synchronous communication.....I think you are thinking in terms of request / response...
But you need only one RFC destination to call RFC function module..which itself will return response as well. So no need of 2 RFC's here.
