How can we compare a program in dev and test client whether there is any difference in the code ?

You will need to set up an RFC destination via SM59, but probably one is already setup.

You can use version management and select remote comparison button to compare two programs in two different systems.
Also you can use transaction SE39 (SplitScreen Editor) as well.
