workflow agent assignemt via organisation unit

any body tell me i want to determine agents assignment for my workflow and that to through organisation structure using rule. in my organisation structure there is only one officer and if he is not there substitute should do the officer work how to create organisation structure for this scenario and i want to create a rule so that mfrnr no. range in between 10 to 100 should use this organisation structure
