We have a requirement to configure ChaRM for 6 system landscape , what should be the approach ? and how to create action profiles if we implement charm for 6 system landscape.

It depends on your system roles and landscape blue print...
Depending up on your system roles ,(DEV,QA,PRE-PROD,PROD,RETROFIT) ,,,,TMS should be configured accordingly logical components are designed.
