In Location Product Substitution R | SAP Help Portal one can find an explanation of the attribute MANDATTRLOCPRODSUBR (Mandatory Attribute Loc Prod Ref).
However, it doesn't seem to explain the actual purpose of this attribute.
If I examine the UNIPA default unified planning area, the attribute is of Data Type NVARCHAR and only has Length: 1
Is it safe to assume that this is a flag? Is the actual use or purpose of this attribute documented somewhere please?

You are perfectly correct!
Another advantage it brings is, while loading the transactional data (like Actual Sales), there might be a combination (example :Customer-Location-Product) not available in relevant master data object (like SOURCECUSTOMER) , the system will reject it, if mandatory attribute is maintained in compound master data object .
In absence of such mandatory attribute, any combination of Customer product Location can be loaded, provided it is maintained in individual master data object. (Product, Customer and Location).

They are mandatory attribute and assigned in planning area as mandatory (Attribute Category).
This concept to link planning object with master data, the moment relevant master data records are deleted it automatically delete all the associated planning object combinations as well.
You will find many examples of mandatory attribute in SAPIBP1.
