Can we connect tenants in two HANA systems using SAP HANA Source System? Is it the fastest way to connect the two tenants?

Yes, see
It makes no difference whether the tenants are part of a single system or part of different systems (SID). You will need to explicitly enable access and direction. Note the section Things to Note About Remote Identities
using SAP HANA Source System
These are not official terms and hence require some explanation. Source system meaning there is replication configured? Using SLT? Data Services? Replication Server? Something else? How does this relate to the first part of the question?

Yes you can do this, simply create two remote sources, one for each tenant you want to point to, the remote sources will have same hostname, but you can use the port to differentiate between the tenants. I hope I've understood your requirement. See my example below of two remote sources, each a tenant on the same host.

As far as I know SDA is part of the core HANA application and there is no parallel SDA middleware. It does however use odbc drivers that are installed on the OS level for various DBs. Here is a nice architecture diagram for SDA/SDI:
