SAP Cloud Connector enables you to connect you on-premise systems to the SAP Cloud Platform (SCP). Considering that your SAP S/4HANA system is hosted on HEC and that you want to allow an application deployed on SCP to have access to it you may use SAP Cloud Connector. You will probably have to talk to your sales representative responsible for your HEC contract to see how you can have SAP Cloud Connector deployed on HEC.
However, if you are talking about using SAP Cloud Connector installed on your LAN to connect to your HEC machines, I believe the answer is no. SCC's configuration takes as connection parameters your SCP landscape/account. This information has nothing to do with a HEC instance. As far as I can tell, a HEC landscape is usually accessible by establishing a VPN tunnel from your LAN to the HEC landscape. Therefore, you should have access to your hosted solution directly from your LAN - it should be transparent to you.
